Introduction
Relenza is an oral inhalant that has been developed by
Glaxo Wellcome to combat the flu. A recent study has demonstrated
Relenza to be effective in preventing the flu, providing an
alternative to the current flu vaccines. Glaxo plans to submit
a separate application for this indication.
How It Works
Relenza inhibits viral
neuraminidase. This enzyme is an essential component in the
replication of both type A and B viruses, strains of
symptomatic influenza. The affected sites of the viral
neuraminidase are conserved so the antiviral action is not
influenced by antigenic changes.
Clinical Tips and Trial
Results
The use of Relenza as a
preventative agent against influenza has some advantages. Once
inhaled, the drug immediately takes effect. Also, Relenza is
not specific to a particular strain of influenza. A possible
disadvantage of the drug is that daily use is required during
the time period when a patient wishes to avoid infection.
A randomized, double-blind,
placebo-controlled study was conducted at two sites during the
flu season. The trial included 1107 patients, and the mean age
was 29 years. The participants were required to inhale 10 mg
of Relenza or placebo per day for a four-week period.
Laboratory-confirmed clinical influenza was defined as
documented infection with two or more of the following
symptoms recorded concomitantly on three or more consecutive
days: cough, headache, sore throat, myalgia, feverishness, or
a temperature above 37.8 deg-C. Relenza was prevented
laboratory-confirmed clinical influenza in 67% (95% confidence
interval [CI], 39%-83%; p-value vs. placebo < 0.001). It
was 84% efficacious (95% CI, 55%-94%; p-value vs. placebo =
0.001) in preventing laboratory- confirmed clinical influenza
with fever. There were no adverse events recorded in the
Relenza group that differed from those in the placebo group.
The results of this study provide evidence that Relenza is
safe and effective in preventing influenza in healthy adults.
